THat depends...
I would DEFINITELY use the block to transmission gasket. Coat the gasket with 518 using your fingers, make it nice and smooth. In most other cases, where you are replacing a thin, paper gasket, you can probably do without. The important thing is whether the assembly requires the spacing of the gasket or not. The oil filter adapter I don't bother with the gasket any more...
