The big hole is simply a place they needed to have an external support as part of the body manufacturing/welding process. Once the body was finished, they just put a rubber plug in there. Try shopping on Restoration Specialties and see if they have a plug the right diameter.
As far as the fuel pump, the hardest part is getting the banjo fitting off that holds the pump in the tank. There is a special tool for that, but usually you get it off by using a hammer and drift pin to hit the tabs to unscrew it.
