I used some Permatex Black Ultra on that weather strip - it sets very permanently once cured, but it doesn't cure fast. I used a piece of oak 1/4" thick 'lattice' about 18 to 20" long (bought at stores like Lowe's or Home Depot) to help force/position the trim into its channel while the window glass was in the door but the rollers were disconnected. I lowered the window halfway before starting, and I replaced the rollers and did exactly what you suggested: I left the window clamping the strip for several hours before I rolled it up. You are wise to do this early. The car I did it on had the strip poorly glued, and it was on the door bottom. Because the lower strip was missing the upper weather strip fell down and was torn by the top rear corner of the window. So I got to buy a new upper trim. The repair is doing nicely.
