If you are careful, replacing the handle won't affect the calibration. Make sure that you note the setting when you remove the handle, and put the new handle on at the same setting. This will ensure that it will torque the same after as it did before. Note however that I'm not saying it will be within tolerance, just that it will read the same as before. If it hasn't been calibrated at least once before, you're flying pretty blind. I have seen plenty of torque wrenches out of tolerance direct from the factory.
