All of the door locks work via the TWICE module that is under the drive seat. The locks will not work if the doors are open and / or there is an issue with the TWICE. The inside lock button should work if you are inside the car with the key on (maybe off too) and all of the doors are closed (no door open indicator on the dash). If it does not then you could have a bad internal key switch.
You have to have a chipped key. It should look like a regular key with a slightly bulging rubber / plastic top. The car will not start without a chipped key that matches / is mated with the TWICE module. If you loose all keys, you have to get a new TWICE module with at least one mated key. This was done to cut down on theft. Now mainly a pain in the butt.
You can run the car without the remote. This year, 2000, had a separate "banana" like remote. In 2001 there was an "integrated" remote with a key's that are a bit bigger on the top with buttons for lock / unlock / trunk unlock. You can order / purchase integrated key shafts for your car from the dealer, purchase used / new "guts" for the integrated keys and then have it mated with the TWICE module. There are tons of information on this on the board.
You REALLY need at least two (2) working keys that start the car. If not and your loose one or if stops working, need to replace the TWICE and keys and have it mated, etc.
Now on to the timing belt.... :-)
