Pull spark plug wires off with the motor running to figure out which cylinder in not running. Pull the plug and replace it if it is in bad condition. I have had the insulator around the center electrode fall down over the gap smothering the spark. A fouled plug will also not fire properly and should be replaced. I would start with a new plug to eliminate that variable. If the cylinder does not come back to life, then start moving backwards to the ignition wires. Swap with a good running cylinder and see if the good running cylinder now becomes the bad cylinder... if it does, then you have found the problem. If that doesn't help, go to the distributor cap and rotor and replace if necessary. If the same dead cylinder does not come back to life, do a compression check. A stuck, bent, or something keeping it open will not allow the cylinder to make compression, therefore will not fire.
