Does the lock cylinder not turn in the lock direction, or does it turn freely and the button just not go down?

On the back of the lock cylinder is a little lever. Turn it one way, and the lever pushes a mechanism that allows the door latch to work, and pushes the button up. The other, and the outside release mechanism (handle button) can't actuate the door release.

If the cylinder turns easily but doesn't lock, I can think of two possibilities. First, the lever is held on the back of the cylinder with a nut. If that nut is backing off, the lever will get floppy. It may still be catching the unlock side, but misses the locking lever.
The other is that the piece that the lever on the back of the cylinder is supposed to hit is just slightly out of alighnment, and it misses. This is the most common one I've run into. Once you get the door panel off and shine a light in, a few actuations of the mechanism show the problem pretty quickly. Usually a little re-bending with a screwdriver or pliers does the trick for another 100K.

If the key won't turn in the lock position, it could still be a loose nut on the back of the cylinder, and the lever is flopping enough to hit something and block movement. A remote possibility is that a burr has developed on one of the tumblers in the lock cylinder, and it just won't turn properly in one direction.

I've run into this before. Chances are you'll spend more time getting the door panel on and off than the time troubleshooting and fixing. It's just a few levers in there. Since the driver's door gets more use than any other door (unless you drive a taxi), stuff in there wears, and the lever method is prone to getting out of alignment.
