Is your wife's car an Automatic? If so there is a Shift Lock Solenoid that makes you depress the brake pedal before you shift the transmission. It sounds like the solenoid is not letting her do that until she stops the car and restarts it.
Possibly the solenoid is acting up and will need replacing. Or the Ignition switch itself is acting up and not sending current to the solenoid.
Here is a test: the next time it happens, have your wife jiggle the ignition switch and press the brakes, instead of turning the car off/on again. If it works it may point at the Ignition switch as the culprit.
