Ah, Barcelona! I lived there for a month during the summer of 2002. My favorite part has to be the old city...but specifically, the part east of Via Laietana, sometimes called La Ribera. It's much less touristy than the Barri Gòtic (although, of course, you should see the cathedral and a few major sites there, as described in all the travel books). The Picasso Museum is in that area, although it's not that great a museum unless you're really interested in the early development of his career...but maybe an excuse to wander through that part of town. Lots of nice cafes there where you can have a casual dinner, too.
It's easy to get Gaudí-ed out, so if you want to see the bare minimum, I'd recommend Casa Milà (aka La Pedrera) and the Sagrada Familia. If you can handle more, they're all described in the books :)
Many guide books gloss over Montjuïc, but it's a wonderful area, especially on a nice day. The art museum there (the MNAC, in the Palau Nacional) has a great collection of art preserved from romanesque and gothic churches. From there, you can wind your way uphill, through the olympic complex, up to the Castell de Montjuïc, from which there are beautiful views of the city and the Mediterranean.
Another great view is from the mountain to the north of the city. I've never been to the top, but there's a bar perched on a cliff about halfway up, and you can sit in the window and look down at the city.
