Road speed, drag coefficient, humidity, air temperature, wind direction and speed, mechanical condition of the car, tire selection and tire pressure, etc, etc.
In a modern car with decent aerodynamics there is probably some advantage to having the windows up and the a/c on at highway speeds. (Not to mention it's quieter and less tiring on a long ride.) But how big an advantage? That'll depend on the car and speed. And if windows down is better, is a mile or so per gallon worth being hotter and less comfortable? And at what speed does the advantage of windows down (or up) go away? Where's the breakeven point? In fact, you might get the best mileage with the a/c off and the windows up. (Try that crossing the Mohave in summer!)
The Mythbuster guys are fun, but they don't always account for all the variables and they make some assumptions in all their tests... and those often go to support the busting of the myth they are working on. And because the only test things with one or two cars the results are projectable to all cars.
A lot of other varaibles make more difference than this. A 30 mph headwind will suck up a lot more MPG than the a/c being on. I've seen the EDU go from 31 mpg to 27 in a 30 mph headwind over 150 miles at 75. (with the a/c pon and the windows up)
