Shimano 105 STI is a very robust shifter. Check for cable stretch - basically increase cable tension by unscrewing the adjuster at the shifter. Next, make sure that the stop limit screw at the derailleur is not stopping the chain from staying on the big ring. Perhaps the limit screw is keeping the pawl at the shifter from engaging.
It's also possible that the entire system is off index by one - increasing cable tension will fix this.
Even with the front ring high-low indexing, it's possible to shift higher than your biggest ring. If you are off index by one, it will seem as if your shifter is not working right - try shifting one lower than your lowest front ring - if the chain drops off, that is what has happened.
