Noel has it right in terms of the two sides.
I've been going down there for my whole life, and enjoy the quaint and the lively.
If you aren't in a hurry, definitely get on 6A in Sandwich and drive along the northern side. Nice twisty roads that take you through a number of little towns. On the other side is Rt 28. All the wicker furniture and beach toys you can imagine. Hyannis and Yarmouth are well populated area's, and happen to be the home of my two favorite places on the Cape. Salty's (cool little lobster shack, home of the twin lobster special and Salty's Brew) and its cheap!
Then a little further East and you hit a bunch of mini-golf places. This to me is a must do on the Cape, and Pirates Cove is by far the best mini....I mean, adventure golf place there is.
I also agree with going to the islands. Martha's Vineyard is a much shorter ferry ride than Nantucket, and both islands are the best of the cape all wrapped up in a little island. Keep in mind that MV really isn't that little, but offers 3 very distict towns.
Hope you enjoy your trip, I miss the cape already.
