There is a local road run here in S. NH for the Crisman brothers (former TSN guys) who died in a car accident, with the proceeds going to an annual high school scholarship. Raises a good chunk of change, IIRC. Another, called the Turkey Trot, raises money for local food pantries.
Connect with a local running or bike club/store to help with logistics. Companies buy some sponsorship and runners pay $X to enter.
For fund raising it's important to get the "feel good" factor out there. You need to be specific about what the donations go towards as this encourages people to give.
Don't forget to do some PR. If you are doing some kind of event, call the editor of your local paper and get him/her to make sure you can get a letter on the op-ed page. If you are having events, make sure the editor knows so he/she can make sure it is covered in the paper.
