Hey Ed,
Well...the number of holes is up to you. You want to do two things- figure out if the mice are in there and also provide a place to spray in some rust inhibitor. There should already be a hole in the front wheel well in the end of the rocker. It should have a rubber plug you can pry out. I guess I have holes in either end of the rocker and probably 3 down the length of it, inside the passenger compartment under the carpet. Every couple of years, I pop the plugs and spray in some stuff.
What do I spray in? Good question. I've toyed with all sorts of things, but have ended up just spraying in new engine oil using a cheap undercoating gun. Seems to work really well. Years ago, I read of a guy that got the idea to FILL his rockers with oil. Leaked all through the carpets and dripped in the driveway for months after!
Of course if you can see in there and find it is full of mouse droppings and nests...you'll have to cut a panel out somewhere to get that stuff out. Maybe a 4 by 4 hole in the wheel well? Then you could snake a shop vac hose in there and get it all clean.
