Anaerobic sealants set up under pressure. That's why the 518(13) is good for the timing cover application. Since the excess that is not captured between the sealing surfaces is not exposed to pressure, it will harmlessly be washed away by the splashing oil.
Robert was right. RTV will set up once exposed to oxygen and the excess can detach inside the engine and circulate into unwanted areas, although in the case of the timing cover, it should drop down and be filtered out by the oil pump pickup screen and filter before it actually gets into the oil passages.
The 518 should be good in the tube for quite some time, provided you keep the cap on it (potential solvent evaporation).
Unfortunately, I don't know the actual chemical structure of the Permatex 518(13), but I'm sure that I could find out if it was of general interest.
