Not necessarily. The oil is more viscous, and operates at higher pressure than the water system does. If it's just a pinhole leak, you'll get enough oil into the water to form a film (it doesn't take much...) in the coolant reservoir, but it would take a pretty large leak to fully mix the two.
If it looks like trans fluid (red) and not the oil you put in the block, then it may be that the trans cooling coil in the radiator is done, and the radiator will need replacing.
