Yes, you can do that, but it does spin the spider gears in the differential faster than they are acustomed to. If these cars had limited slip differentails then it would definitely be a bad idea. Also, if you have TCS, the car will apply the brake on the spinning wheel because it has no traction.
A lot of times the test is ineffective in isolating a noise because there is no load on the wheel bearing and CV joint, which makes a difference.
