1. Drive in 6th gear with speed maintained by cruise control at a setting you feel is right. At a fixed point (Eg. mile marker) approaching the incline, CLEAR the computer. At another fixed point (perhaps another mile marker) after the incline, observe your mpg.
2. Repeat test in 5th gear (this should be the only variable). Speed and the fixed points used should be the same.
Try this comparison on 2 different days wherein the temperature and humidity is very similar. Large differences in these factors will affect the results. Finally, ensure that the load you're carrying is the same (4 people one day and you alone on the other day will really have an effect. Also, note that gasoline weighs about 6 lbs per gallon, so try to have a similar amount of gas in the tank.
You may want to perform this test several times on the incline you have in mind. Perhaps this test approach could be used over a greater portion of your commute.
