One thing to watch out for is there are a lot of counterfeit flash drives around. The package might say they're 128GB or 256GB or more and the drive's firmware will even have been hacked to report that bogus capacity to the operating system, but are actually the cheapest possible 8GB drives inside. Same goes for SD and MicroSD cards. I know people who have been snagged by that scam. Try to buy only from well-known, reputable vendors but sometimes even they will unknowingly get a shipment of bad drives. At least with a reputable seller you can get a refund if that happens.
USB2 is more than sufficient for audio. Best arrangement of the files depends on the player. If it has a true file manager you can lay out the directories and files as you please. Some however will insist on trying to categorize the files by a metadata and/or filename or have other limitations. (We found for example on the wife's Subaru Outback that the radio's USB player does not grok the concept of subdirectories in its interface.) This won't be a problem if mainly playing mp3 files on a computer.
