Assuming you are going to see the scenery (if you are going for the beer, just go to the Rocks in downtown Sydney and do a pub crawl & forget the car). This is perhaps biased as I am an ex-Victorian. Driving from Sydney to Melbourne & return (or vice versa) - take the inland route one way (Rte 31) and the coastal the other (Rte 1). In Melbourne, visit the Healesville Wild Life Sanctuary (about 40 mls) (has platypus, koala, kangaroo & more) with some good vineyards/restaurants in vicinity. Visit Ballarat/Bendigo (major towns in the Aussie gold rush) & especially Sovereign Hill - a museum village of the gold rush. Take the Great Ocean Rd - starts beyond Geelong 50 mile SW of Melbourne and about 150+ ml drive to Warrnambool (allow a full day) then to Grampians (mountain range) & abck to Melbourne next day. The mountains NE of Melbourne. If you like hiking, Wilsons Promontory SE of Melbourne, Penguins at Philip Island.
Near Sydney visit the Blue Mountains and for best race track in the world, to far side of the Blue Mts to Bathurst (track is probably in use at Easter but can be driven most times). Take the ferry ride from The Rocks to Manly. Drive along coast from Manly to Palm Beach (not on weekends). I'm sure the people from Sydney will give you more ideas.
