I think you will need to lock the flywheel to try the leakdown. You don't want he engine moving in reverse when the cylinder is under pressure, or forward for that matter....
I bought a flywheel lock that bolts on to keep it from moving. There are other ways of course, but I usually have to work alone, and having the bolt on lock helped.
Also, I just look at the distributor to get a rough idea where #1 is, and then make the finer adjustment, remembering not to rotate the engine in reverse. There is a mark on the block that is the best visual reference point, and of course the line in the plastic bell housing that is probably within a city block of being right.
