If you are not aware, it is advisable to remove all of the spark plugs while setting up the timing chain. As Landjet notes, you will want to rotate the engine through at least a couple of turns by hand to make sure everything is spot on. It is much easier and a smoother process if the spark plugs are out and you are not fighting with the compression of the cylinders.
