I used to do the 380 mile run from home in NH to Webster, NY 4-6 times a year. I'd usually get better mileage coming back than going out, mostly due to the headwind on the NY Thruway heading west. You don't necessarily notice a steady 15 mph headwind when driving, but the car does. On one very windy day I watched my indicated mileage (which runs about 1.5 mpg high) drop from almost 29 to around 26.5 when battling a 30 mph headwind west of Syracuse.
How you go up hills, use of cruise control, whether it's dry or wet out, and how often one dives into the boost all add up to real differences.
I'd also find I get slightly better mileage on mid-grade than premium, although that varied by brand of premium. The Irving I'd buy in NH seemed to do better than the Irving I'd buy in NY. And Hess gas in NY always delivered worse mileage than other brands.
