I normally use Nitrile gloves and they've been fine. The most important thing really is eye protection. Be sure to wear safety goggles that are designed to protect against chemical splashes. These will have some kind of baffles on the air vents to prevent liquids from entering. You need to wear them tightly enough to seal properly -- you absolutely do not want to have pressurized refrigerant, oil, or flushing chemicals spraying in your eyes if there is a mishap.
As far as evacuating the system, in a public forum all I'm going to say is that it is illegal to deliberately vent R134a to atmosphere. (Though frankly it will all ultimately wind up in the atmosphere at some point due to leaks and accidents.) What happens in your own garage is of course your own business.
Now if you asked that question and happened to be using an alternative refrigerant such as propane which has no restrictions on atmospheric venting, I would say to simply connect your gauge set and slowly open the valves to bleed off any pressure in the system. Then do your repair work, pull vacuum, and recharge.
