Refrigeration is the safest method, but it does not kill the bad bacteria, merely slows it's growth. By the time the chicken is defrosted, the level of toxins is low enough to avoid causing illness.
Defrosting at room temperature allows bacteria growth to accelerate, at least on the surface, to a level at which some people might become ill. I say "some people", because apparently your mother-in-law and her family are resistant. This can either be due to genetics, or the fact that her family became immune due to repeated exposure to the toxins.
