In my experience McAfee is pretty much bottom-of-the barrel, I'm always seeing problems with it. If your system is not too far gone I would try to download and use Combofix, then use Malwarebytes to mop up. (You may need to boot into "safe mode with networking.") If the infestation is really deeply rooted you may need to reload Windows using your system recovery discs (or recovery partition). If you do this, back up any important data first.
That's pretty much the same drill with any operating system, following a major security breach the only way you can be sure the problem is gone is by performing a clean install.
Once you get this problem fixed you may want to consider better antivirus protection. Everyone seems to have their favorite. On the Windows systems that I deal with I've seen the fewest virus problems on computers running Kaspersky Antivirus. (I don't care for full-blown "internet security suites" as they are very invasive and can cause severe performance and stability problems, stick with a good, solid antivirus/antispyware package.)
