Well, English will NOT be a problem in Amsterdam. I've been there 4 or 5 times, and it won't be a problem at all.
Don't rent a car. Even if you plan side trips, the train system is wonderful. I suggest Brugge, Belgium as a romantic side trip.
Coffee shops will be pretty simple to find. Airport to Centraal Station on the train, walk out the front onto the Damrak, and follow your nose. I've never been into "smoking", but I've hear the most touristy (therefore annoying) coffeeshops are near the station. Head back to the Leiseplein area.
As for things to see, the art is wonderful, shopping is world class, the orchestra is also great. Check out the Van Gogh museum and the modern art museum next door. There's the Anne Frank Huis. If you're a jazz fan at all, look up the schedule for the Bimhuis. It's even with NYC's Knitting Factory for quality of cutting edge artists and sound.
Since you're married (and a woman), the Red Light District is probably not the ideal destination. But take a stroll through, it's truly amazing.
If you don't want to look like an american, just roll with the small beer glasses. You can order a large, but you'll stick out as a tourist. If you're into the world's finest beers, there's a neat little Belgian bar called Café Belgique at Gravenstraat 2.
