Be on the lookout for how it feels both in fit as well as how it feels on rough roads. Also get a feel for the type of frame you want; aluminum, steel, carbon, titanium, or any combo thereof.
100 mi can be torture on some aluminum bikes but Ok on others. As a larger rider, aluminum may be OK for you and it won't rust over time byt may crack at the welds. Carbon is great as it is smooth, stiff, and light but it's possible that one bad crash could hurt the frame pretty badly. Steel is great but it's typically heavier and can rust and looks less sexy to some. I personally like steel if done right. Some of the nicer lighter steel bikes are as harsh as aluminum though. Titanium combines the benefits of steel with aluminum, in my humble opinion, but I liked carbon better. Since I'm only 5' 8'', the smaller frames make aluminum even worse for me. My favorite aluminum bike was the Cannondale though. Nice ride.
For components I assume that you want to be value oriented so Shimano is probably a good idea. I suggest 105 or better. If you want to treat yourself you can go with Campagnolo but you have to pay twice as much for the same thing.
How about checking out a Bianchi Veloce triple? It's a tad heavy but great for longer rides. Bicycling magazine recently had a buyers guide. Might want to see if you can find that one. It was some time between Feb and now.
